dc.description.abstractМелатонинът е хормон с широк спектър на биологична активност, но ролята му в процесите на хемостазата е недобре проучена. Целта на дисертационния труд е да се проучи влиянието на мелатонина върху пусковия механизъм на хемостазата у плъхове. Изследват се ефекта на мелатонина и инхибитора на неговите рецептори - лузиндол върху интегрални показатели на хемостазата; броя и функционалната активност на тромбоцитите; плазмения фактор на фон Вилебранд; тъканния фактор и инхибитора на пътя на тъканния фактор. Използвани са 156 мъжки плъха Wistar, отглеждани при естествен режим светло/тъмно 12:12 часа. Дневната доза мелатонин от 0.2 mg/kg b.w. и лузиндол 0.4 mg/kg b.w. бяха приложени s.c. два пъти дневно, през 12 часа, в три последователни дни. Плъховете бяха разделени в четири групи: първа група - инжектирани с мелатонин, втора група - с лузиндол, трета група - с лузиндол и един час по-късно с мелатонин, четвърта група - контролна, с физиологичен разтвор, разтворител на хормоните. Резултатите сочат, че мелатонинът предизвиква значимо скъсяване на аРТТ, РТ и ТТ. Лузиндолът, приложен самостоятелно и в комбинация с мелатонин, силно удължава аРТТ, РТ и ТТ. Мелатонинът увеличава плазмената концентрация на β-TG и PF4, а самостоятелно приложеният лузиндол и претретирането с лузиндол редуцира значимо нивата на тези показатели. Епифизният хормон значимо увеличава броя на тромбоцитите, плазменото ниво на vWF:Ag и активността на vWF, а лузиндолът (самостоятелно и в комбинация с мелатонин) ги понижава. Мелатонинът повишава плазмената концентрация на TF и същевременно понижава плазмените нива на свободния TFPI:Ag и активност на свободния TFPI. Лузиндолът, приложен самостоятелно и в комбинация с мелатонин, намалява плазменото ниво на TF и повишава нивото и активността на свободния TFPI.BG
dc.description.abstractMelatonin is a hormone with a broad spectrum of biological activity, but its role in the processes of hemostasis has been insufficiently explored. The purpose of this disseration is to study melatonin's influence on the triggering mechanism of hemostasis in rats. The examination spans the effects of melatonin and its receptors' inhibitor - luzindole - on integral hemostatic indicators; thrombocyte number and functional activity; plasma von Willebrand factor; tissue factor and inhibitor of the tissue factor pathway. 156 male Wistar rats were used, kept at a natural regimen of light/dark 12:12 hours. The daily doses of melatonin 0.2 mg/kg b.w. and luzindole 0.4 mg/kg b.w. were administered s.c. twice a day, at 12 hour intervals, over the course of 3 consecutive days. The rats were split into 4 groups: first group - injected with melatonin; second - injected with luzindole; third - injected with luzindole and an hour later with melatonin; fourth - control, injected with physiological solution - hormone solvent. The results indicate that melatonin induces a significant decrease in aPTT, PT, and TT. Luzindole, applied separately, and in conjunction with melatonin, strongly increases aPTT, PT, and TT. Melatonin increases the plasma concentration of β-TG and PF4, while the separate application of luzindole and luzindole pretreatment markedly reduce the levels of these indicators. The epiphyseal hormone significantly increases platelet count, plasma levels of vWF:Ag, and activity of vWF, while luzindole (applied separately and in conjunction with melatonin) decreases them. Melatonin increases the plasma concentration of TF while lowering the plasma levels of free TFPI:Ag and the activity of free TFPI. Luzindole, applied separately and with melatonin, decreases the plasma level of TF and increases the level and activity of free TFPI.en_US
